No. Only one controller has to be signed in with the CC membership while your game is running - the others can be not signed in, local, LIVE, any type of account.
In fact common fail reasons are not checking purchase privileges, since to buy a game you have to be signed in with an account that does not have purchase restrictions. So, it's good test your game with varying combinations of not signed in / local profile signed in controllers.
